Stem Cell Sequencing/Epigenomics Analysis

Michael Buck, PhD (right), directs the Stem Cell Sequencing/Epigenomics Analysis facility and uses its resources to collaborate on studies with Yijun Sun, PhD, and other UB researchers.

We are a resource for epigenomic analysis of stem cells. Our facility functions interactively with other stem cell facilities at UB to characterize stem cells and their differentiated progeny.

The goals of this facility are to:

  • characterize the transcriptome and epigenome of all stem cell types and their derived lineages
  • provide experimental and computational support for investigator-initiated genomic expression or epigenetic experiments

This facility can determine:

  • gene expression and microRNA expression profiles using RNA sequencing
  • genomic DNA methylation using methylated DNA immunoprecipitation and sequencing
  • genome-wide histone modifications and transcription factor binding site localization using ChIP-seq
  • nucleosome positioning using gel-excised mono-nucleosomes and high-throughput DNA sequencing
  • chromatin accessibility by FAIRE-seq

Instrumentation

Our facility has instruments including:

  • illumina HiSeq 2500, a flexible and scalable platform that features two run modes, rapid run and high output run mode and the ability to process one or two flow cells simultaneously
  • illumina MiSeq, a platform specializing in more targeted sequencing geared toward smaller genomes and smaller search space investigation

This core facility is one of four stem cell research facilities maintained by WNYSTEM.
